Under the guidance of Dr. Cletus J M D’Souza, Professor, Department of Studies in Biochemistry, University of Mysore, Manasagangotri. Mysore – 570006. Aim and scope of the project work : Since the phospholipids and fatty acids play a major role in atherosclerosis, the main objective of this study was to characterized the changes taking place in the fatty acid composition of oxidized LDL. The fatty acid composition of phospholipids is greatly influenced by the dietary fatty acids. Hence we wanted to characterized the nature of fatty acids in the LDL particle and see what happens to these fatty acids on oxidation with Cu++ ions. Objective of the study : The overall objective of the study was to study the changes taking place in the lipids and proteins of LDL particle on oxidation ,specific objective are : 1. To prepare LDL from human serum by ultracentrifugation. 2. To oxidize LDL by Cu++ ions and to characterize the changes in proteins and lipids. 3. To standardize a simple assay for PLA2 and to test the stimulation of endogenous PLA2 by oxidation. It is a condition that cause inflammation and accumulation of fat and fibrous tissue in the liver. NASH occurs in those who drink little to no alcohol. NASH has been observed for serious liver destruction and enhanced liver mortality. It has systemic consequences concerning insulin resistance, metabolic complications and cardiovascular diseases.
