ACADAMIC PROJECT TITLE
Impact of UV-B radiation on the siderophore production of the Rhizobium isolates and their characterization using Bioinformatics tools.

Summary
The impact of ultraviolet B radiation on the siderophore production of Rhizobium isolates was done. Initially, the isolation and characterization of Rhizobium was done. They revealed that the 5 different plants of Arachis hypogeae harbored 5 types of Rhizobium strains. This was confirmed by their characters such as exopolysaccharides production, motility, acid production, nitrogen content, ammonia content, nitrite excretion, siderophore production and antibiotic sensitivity tests. In all these biochemical aspects they greatly differed from each other. The growth curve also showed a greater variation between each isolates.
Each isolate produced their own siderophore and they were recovered in the form of powder. The ability to produce siderophore varied with different strains which were proved by their quantity and color.
On mutating the Rhizobium isolates using ultraviolet B radiation with different time of exposure, they had an impact over siderophore production in all the isolates. By comparing these results it showed that in Rhizobium itself they produced different type of siderophores. This was also confirmed using Bioinformatics tools. The multiple sequence alignment and the phylogenetic tree proved that each Rhizobium strain produced different kind of siderophore. They varied in their protein sequence. The phylodraw also revealed that the siderophore of each Rhizobium strains had an evolutionary relationship with each other.
