Summer Training, Natco Research Centre Private Limited, Hyderabad.
Description : Docking was done for HIV-1 reverse transcriptase protein and using that parameters we docked five FDA approved drugs and other 10 molecules having structure similar to the approved drugs. Two among the 10 molecules gave best result compared to FDA approved drugs. The algorithm used in Auto Dock Vina is “Genetic Algorithm”.
(2).Virtual screening of Protein Tyrosine Phosphatase 1B with Zinc Database : An approach for QSAR properties and Pharmacophore group Design
Five months project in Silicon Bioinformatics (A unit of Muvva Biosolutions Pvt.Ltd), Hyderabad.
Description : PTP1B is encoded by PTPN1 gene. It plays a major role in type 2 diabetes. Structure of PTPN1 gene is modeled using MOE and virtual screening is done on 47,000 ligands downloaded from ZINC Database. Based on dock score 20 analogues are predicted as best and 16 analogues satisfy Lipinski rules. For these 16 analogues hydrogen bond interactions, QSAR properties and Pharmacophore groups are predicted using PyMOL, HyperChem and LigandScout softwares. Finally, 7 analogues are identified as best inhibitors for PTP1B. The scope of the study extends to use these 7 analogues as leads for drug discovery.
