Project Title : Combined Effect of Dietary Phytochemicals and Chemotherapeutic
Agents on Cervical Cancer Cell Line(HeLa 60)
Abstract :
Combinational therapy refers to the use of two or more kinds of chemotherapeutic drugs. Single modality drug treatment may be more effective at a certain stage of cancer which alone may not affect the tumor completely. The rationale for combinational therapy is that effect of a single drug may be enhanced by the inclusion of another drug that might act additively or synergistically. An advantage of including dietary phytochemicals in the cancer treatment regime is that they have apparent benefit in minimizing the toxicity and chemoresistance and at the same time alleviate the patients from the possible side effects of chemotherapeutic agents. In the present work, the dietary phytochemicals used are vitamin c, indole-3-carbinol(I3C) and the chemotherapeutics includes sulidac,an active NSAID. Results from this study showed that sulindac alone and in combination with dietary phytochemicals reduced the number of cancer cells significantly. But sulindac when used alone seemed to kill the cells by necrosis. The same sulindac when combined with phytochemicals produced similar amount of cell death, however the mode of cell death was preferably by apoptosis as evident from chromatin condensation and DNA fragmentation. Hence, the combination of sulindac with different dietary phytochemicals should be explored in future for understanding its mechanism of action at the molecular level.
2. SUMMER PROJECT(2010) : Duration- 2 months

Project Title : Development of Mammalian Double Gene Expression Vector for Antibody
Production.
Abstract :
Recombinant vectors are valuable tools in the biopharmaceutical industry with a number of novel vectors being developed every day. Antibodies are complex proteins with two polypeptide chains. These are often expressed in mammalian cells by co-transfecting light chain and heavy chain containing plasmids. However, co-transfection can lead to variable in the copy number of both heavy chain and light chain, there by affecting the protein productivity. Double gene expression vectors can overcome these problems. The double gene expression vector contains two independent transcriptional units. In the current design The first transcriptional unit comprises sequences for the CMV promoter, multiple cloning site (MCS) and a Polyadenylation Sequence and the second transcriptional unit comprises sequences for the SV40 promoter, MCS and polyadenylation sequence. The double gene expression vector (pUB-C-S) was constructed by ligating the BglII and BamHI fragment (974bp) form pSI plasmid with BglII digested pCI plasmid. The presence of two independent transcriptional units in the recombinant plasmid was confirmed by colony PCR.
