The intra islets precursor cells gave rise to patches of well organized polygonal epithelial like cells within 4 days of the addition of serum free media. The purity of the islets was assessed by Dithiazone staining, culture islets were fixed for immunoassaying of insulin, glucagons, somatostatin and epithial cell marker cytokeratin (CK-7 and CK-19) was done. Insulin secretion assay of islets was carried out with 5.5 mM glucose and then 16.5 mM glucose of l hour as a stationary set up. Western blot analysis, RT-PCR analysis of insulin, PDX-1, TNF Alpha, Glut-2 IRS-1, Sox-2, Nanog and Oct-4 was carried out.
