Final Study project : Study of wood degrading microorganisms.

Overview : The study focused on the isolation, characterization, degradation and enzyme activity of different strains of bacteria and fungus from decayed wood sample. Serial dilutions prepared from the degraded wood were selected for enumeration of bacteria and fungi. A total of 12 microbial species were isolated from the decaying wood; 4 bacteria and 8 fungi. Evidence for the microbial utilization of cellulase was detected by hexadecyltrimethyl ammonium bromide with clear zone around the growth. Pectin utilization was detected by flooding the culture plates with freshly prepared Iodine-Potassium iodide solution. Among 3 fungi; brown rot fungus, white rot fungus, soft rot fungus; brown rot fungus shown maximum cellulase degrading activity upon other 2 fungus is 9µg/ml/min. After 48hrs of incubation,Aspergillus sps showed maximum pectinase activity(10.13µg/ml/hr) compared to 12, 24, 36hrs. The amount of reducing sugar liberated was quantified using glucose as standard by DNSA method. The purpose of the study was to isolate microorganism which can produce partially purified cellulase and pectinolytic enzyme have potential commercial values.
Study project : Isolation and Characterization of Rhizobium from Mimosa pudica and Trigonella faenumgracum.

Overview : In the present study, morphological and biochemical characterization, their genomic and plasmid DNA analysis were conducted to characterize Rhizobium species isolated from Trigonella faenumgraecum and Mimosa pudica. Bergey’s manual was followed for morphological and biochemical studies. At 28 and 35°C, lower concentrations of NaCl (0.1 and 1.0%) favored growth of MSSP, while 2% NaCl extended the lag phase. Rhizobium isolated from Trigonella faenumgraecum has responded to biotin, thiamine and its combinations. Results indicate that the Rhizobium isolated from Mimosa pudica found to be similar to MSSP strain of Burkholderia species.
