BCIL TRAINING PROJECT :
TITLE : Isolation of variable region from Human immunoglobulins.
Description :
The study was done to isolate variable region, VR from the FAB region, FAB isolated from papain digested immunoglobulins. Separation of FAB was done by developing a cost effective Hydroxyapatite column. The end goal of this project is to exploit the fact that the antigen binding fragment binds to epitope of an antigen to mediate an immune response; in other words VR facilitates antibody therapy. The fragments of antibody can be employed also in diagnostic procedures like ELISA and in manufacturing of Diagnostic kits.
Roles and responsibilities :
- Human serum was collected.
- Capryllic acid treatment was done to precipitate the proteins except immunoglobulins.
- IgG was purified by Protein-G Affinity chromatography.
- IgG was digested using papain enzyme to generate FAB.
- FAB was purified by Hydroxyapatite chromatography (Pseudo-affinity).
- FAB samples were concentrated by Sucrose gradient.
- Protein concentration was estimated by Lowry’s Protein estimation.
- Purified FAB was further digested to isolate variable region.
- SDS-PAGE was performed to check the purity of protein.
