    Research Summary :
First Asymmetric Syntheses of Permethylated Caffeic Acids as Anti-HIV Agents. This part of
thesis developed a route for the asymmetric synthesis of methyl lactate and its antipode. Application
of the general protocol culminated in the asymmetric syntheses of both the antipodes of
permethylated rosmarinic acid, salvianolic acid D, salvianolic acid C, salvianolic acid A, and their
biological activity against HIV-1 virus replication was evaluated. Among them, permethylated (+)-
rosmarinic acid and (±)-salvianolic acid C were found to inhibit the replication of HIV-1RTMF (AZTresistant)
virus with IC50 values of 34 µM and 50 µM respectively.

Synthesis of Heptamethyl Lithospermic Acid. Herein, we have established the two different
routes for the synthesis heptamethyl lithospermic acid. In the first strategy, a modular, flexible, and
convergent route to the heptamethyl lithospermic acid along with its diastereomeric form has been
accomplished from 3,4-dimethoxybenzaldehyde in 11 steps. Second strategy, discloses the
alternative method for the synthesis of trans-cinnamic acid core of lithospermic acid and also
successfully applied for the synthesis of heptamethyl lithospermic acid. This synthetic route can be
applicable for asymmetric synthesis of heptamethyl lithospermic acid.

Total Synthesis of Rosmarinic Acid and (R)-Oresbiusin A and B. This part of thesis presents the
total synthesis of polyphenolic acids which exploits tert-butyldimethylsilyl (SiMe2t-Bu) as an ideal
protecting group. Total synthesis of (±)-rosmarinic acid was accomplished in 7 synthetic steps from
3,4-dihydroxycinnamic acid with 42% overall yield. In addition, developed an efficient, convenient
route for the total synthesis of (R)-oresbiusin B through a linear approach from 3,4-
dihydroxycinnamic acid in 6 steps with 64% overall yield. Furthermore, efficient and flexible routes
were established for the syntheses of orthogonally protected skeleton of salvianolic acid C and
    salvianolic acid D.
