EFFECT OF UV RADIATION ON ZEBRA FISH
Description :-
The zebra fish (Danio rerio) contributes to the identification and characterization of genes and pathways involved in development, organ function, behavior and disease. UV radiations are commonly used for Therapeutic applications, for example cancer treatment at lower dosages, waste water treatment, and used in laboratory purposes. UV radiation is used in this study for mutating the species, because it has shorter wave length and higher intensity. The UV radiation invades the cells in the early embryonic stages, namely the four cell stage. It is established that these blastomeres are the precursors for various cell lineages that make up organ systems, namely the nervous system and circulatory system in mature larvae .The zebrafish embryos have been treated with the various time intervals (20s, 40s, 60s, 80s, and 100s) of UV radiation, and the intensity of the radiation is 1.253 watts/cm^2. Twenty four hours post-fertilization embryos were dechorionated and incubated for 90 mins in colchicine to arrest cells in metaphase. Colchcine is a metaphase inducing compound which arrests the cell division in late metaphase stage of cell division.
Chromosomal aberrations of the embryos exposure by UV radiation was seen by arresting the cells in metaphase and the chromosome spreads were viewed
followed by photography in phase contrast microscope. Significant chromosomal aberrations are maximum in those embryos which were exposing to UV radiation for 100 seconds. More time exposure of UV radiation increase in mortality rate and chromosomal aberrations along with decrease in heart beat rate were observed. The time intervals of UV radiation ranging from 0-20 seconds were observed as the minimum lethal exposure in the case of zebrafish embryos.
Roles and Responsibility :-
Obtained the final result from chromosomal abberiation of Zebrafish by the embryos exposure by UV radiation was seen by arresting the cells in metaphase and the chromosome spreads were viewed followed by photography in phase contrast microscope
