    ACADEMIC PROJECT (M. Pharm.) :
    PROJEC TTITLE : In- vitro and in-vivo pharmacokinetic drug drug interactions study of Tolbutamide with Atorvastatin at GVK Bio Sciences Pvt Limited, Hyderabad.
    DESCRIPTION : Patients with the metabolic syndrome and/or Type 2 diabetes mellitus continue to have a high risk of coronary heart disease (CHD) and progression of atherosclerotic lesions. Tolbutamide, a first generation sulfonylurea, is preferred in therapy because of its selective inhibitory activity toward pancreatic K+ATP channels, low incidence of producing severe hypoglycaemia, and other haemobiological effects. Antihyperlipidemic drugs like statins are widely used for prophylactic treatment in dyslipidemia and atherosclerosis. Among statins, HMG CoA reductase inhibitor, Atorvastatin is well established and widely used because it is indicated for primary hypercholesterolemia, mixed dyslipidemia, atherosclerosis, hypertriglyceridemia, and dysbetalipoproteinemia. Atorvastatin is metabolized by P 450 CYP 3A4 isozymes in the liver and it also has moderate inhibition on metabolizing enzymes P 450 CYP 2C9, CYP 2D6. Hence, there is more possibility of Atorvastatin for inhibition of metabolism of tolbutamide, which is metabolized by CYP 2C9 By using pooled rat liver microsomes in-vitro and by analyzing in-vivo rat plasma samples using LC-MS/MS, present study is planned to predict the pharmacokinetic drug interactions if exists.
