    Title “The role of nitric oxide and curcumin in mediating vasorelaxation in Goat Ruminal Artery”Under the guidance of Major advisor, Dr S.C Parija, Ph.D(I.V.R.I), P.D.F(UK), Head, Dept. of pharmacology and toxicology, OVC,OUAT
    Description NO is generated in biological tissues by specific nitric oxide synthases (NOS) i.e eNOS/nNOS/iNOS which has t1/2 of a few seconds in aqueous media with a low oxygen concentration, and is soluble in a lipid-rich environment. NO has been reported as an important regulator and mediator of numerous processes in the nervous, immune and CVS, including smooth muscle relaxation thus resulting in vasodilation of the artery and increasing blood flow, neurotransmission and has been associated with neuronal activity and various functions like avoidance learning, macrophage mediated cytotoxicity for microbes and tumor cells. Currently, exogenous NO sources constitute a powerful way to supplement NO when the body can not generate enough for normal biological functions.
    Result We have conducted Isolated tissue response to various ligands of NO pathway to study the drug receptor interaction further supported by Molecular Biology assays like 1-D Gel electrophoresis, 2-D gel electrophoresis, immunobloting etc. to study c-GMP dependent NO pathway in GRA.
    We have investigated involvement of c-GMP dependent NO pathway in vasorelaxation of GRA which may be e-NOS independent. All the contraction studies were carried out with four chambered automatic organ bath (Panlab S.I, Spain) and Power lab data acquisition system (AD Instruments, Australia) with Lab chat6 Pro software. All the analysis were carried out using GraphPad Prism (GraphPad Prism Software, San Diago,CA,USA).The proteins were analyzed by 1-D GE(one dimensional gel electrophoresis) in a mini-protean 3 electrophoresis cell(Bio-Rad Laboratories, Richmond, CA) using 5%(w/v) staking gel and 12% separating gel which separated the extract into ~51 protein bands in molecular range of 6-205kDa. 2-D GE was done by carrying out IEF (Iso electric focusing) in a Protean IEF Cell using 11 cm IPG strips (pH 5-8) followed by SDS-PAGE in a Protean II xi Cell. Coomassie- silver double staining separated the soluble ruminal artery vessel proteins into ~200 spots. The immunoblot analysis of the vascular protein extracts from four untreated group identified hsp90 which is a co-stimulator of eNOS.
