Abstract : Hplc is a sophisticate technique; it is also evident with the results drawn from two popular methods of quantification that is spectrometric and hplc shown same results matching with other very less error. However sensitivity is very high with hplc and results are reproducible with using hplc in quantitative analysis.hplc show reliable and effective as well as efficient method in determining quantities and concentrations of plant pigment like carotene.

Abstract :
The Cucumber (Cucumis sativus) is a widely cultivated plant in the gourd family Cucurbitaceae, which includes squash, and in the same genus as the muskmelon. Cucumber seeds possess similar properties to those of the allied Pumpkin (Cucurbita Pepo, Linn.) which are distinctly diuretic, but mainly employed as a very efficient taeniacide. The emulsion made by bruising Cucumber seeds and rubbing them up with water was formerly thought to possess considerable virtue and was much used in catarrhal affections and diseases of the bowels and urinary passages.
The process of in-vitro propagation commenced with the germination of seeds. The seeds were surface sterilized in Tween-80 and then in 0.1N HgCl2 for 10 minutes and washed with distilled water. The sterilized seeds were then inoculated onto a sterile paper hood placed in sterile boiling tubes containing autoclaved water. 5 days old in-vitro raised seedlings produced green nodular leaves. They were surface sterilized with 0.1N HgCl2 for 15 seconds and then washed with distilled water. Then they were transferred onto MS medium containing IAA (0.05 – 0.1 mg/litre). It resulted in the callus induction in 7 days. Adventitious shoots were produced from the organogenic callus when it was transferred to MS medium supplemented with IAA in different concentrations ranging from 0.1 – 0.5mg/litre. The shoots were excised from the callus and transferred to MS media supplemented with higher concentrations of auxins for the root induction. Thus the MS media supplemented with different concentrations of auxins yielded organogenesis from the callus developed from the seed explant.
