TITLE : Bio-control of stem rot in ground nut incited by Sclerotium rolfsii and
Invitro compatibility of potential native antagonists with fungicides
Abstract :
Groundnut is an important oil seed crop of tropics and sub-tropical regions of the world. Among the fungal diseases, Stem rot of groundnut caused by Sclerotium rolfsii is potentially very destructive disease. A detailed study on isolation of pathogen, rhizosphere mycoflora, antagonism of rhizosphere against S.rolfsii and integrated management of the disease with biocontrol agent. Eight different mycoflora were Aspergillus flavus, Rhizopus sp, Pencillium, A. niger and Trichoderma isolates (TGI, TG2, TG3, TG4) isolated consistently from the rhizosphere of groundnut. Among them TG2 isolate was found to be significantly superior over others in inhibiting the mycelial growth of S.rolfsii to the extent of 67.83%.

Research summary :
Abstract
Phytase is an important industrial enzyme hydrolyzing phytate to release inorganic phosphate. A microorganism with thermostable Phytase activity was screened from a total of 17 isolates from the gut of Eisenia foetida were collected from top ploughed horizon (0-20 cm) of vermicompost pit from and around Tirupati region, phytase producing microorganisms were selected by size of clear zone on the plate of Modified phytase screening medium (MPSM) and their phytase activities were measured by Trypti soya liquid assay method. An isolate PSB-1 was finally selected among some bacteria with a high phytase activity by determining thermo stability of phytase. The isolate was identified as Bacillus Subtilis MTCC 9878 by morphological observation, biochemical test, and 16s rRNA similarity analysis. The optimum activity of crude phytase Bacillus Subtilis PSB-1 was observed at 700c and pH 7.0 .The residual activity of the phytase was over 78% after heat pretreatment at 800c for 10 min, which indicates that this enzyme was to be highly thermostable .The crude phytase was also stable at broad range from 3.5-9.5 PSB-1 was the most efficient strain depending on its capability to solubilize P, utilizing a wide range of carbon, nitrogen sources and at different pH, temperatures.
Phytase production is carried out in phytase screening media supplemented with 2mM phytate per liter the culture broth was clarified by centrifugation, proteins were precipitated by adding 3 volumes of cold ethanol(-200c) and precipitate was redissolved in 100mM Tris –Hcl (pH7.5)supplemented with 1mM cacl2 and assayed for phytase activity as described by shimizu. One unit of phytase was defined as the amount of enzyme required to liberate 1 micromole of inorganic phosphate per minute.
And the enzyme is purified by 45%, 65% and 85% of ammonium sulphate fractionation and passing through PD-10 gel filtration column (Pharmacia)in to a 100Mm Tris-HCL buffer pH-7.5 and the molecular weight was determined by SDS-PAGE i.e.,46 kDa .
Earthworm is a unique habitat of its kind, was investigated as a source of bacteria to screen for the production of novel enzymes. Soil experiments were conducted by treatments as follows : (i)Untreated control, (ii) sodium phytate (sp) - 30 Kg P ha-1., (iii) Farm yard manure (FYM) – 40 Kg P ha-1, (iv) (FYM) – 40 Kg P ha-1 + (SP) - 30 Kg P ha-1. Plant growth promotion influenced by inoculation with phytate solubilizer was measured in terms of ground nut plant height and fresh weight and root length over uninoculated control after 15 days of germination results in the present study showed the efficiency of this isolate to make available P from sodium phytate in presence of farm yard manure. Plant growth promotion of ground nut was not increased significantly compared to controls.
Phytate solubilization is a complex process involving both organism and soil, needs more attention between laboratory methods to soil tests and seed inoculation experiments. Microbial phytases can be exploited for efficient conversions of phytate to phosphates when given as green manure to the soil for improved yield.
The present study concludes the gut flora of earthworms contribute phosphates to crops when supplemented to soil to replace the chemical fertilizer Phytase producing bacteria are substrate specific. They show greater resistant to proteolysis and better catalytic efficiency. Phytase has broad applications in biotechnology, therapeutics, agriculture and in reducing environmental pollution.
