    Project details :
    1) Project name : Partial isolation and characterization of acid phosphatase from the leaves of Allamanda cathartica L.
I have done this project work under the guidance of Mrs.C.N.Shanthi Vasudevan lecturer in Biotechnology, Department of Botany, St.Berchmans College, Changanacherry.

Fifty two plants were screened for the enzyme acid phosphatase and Allamanda cathartica L was selected having comparatively higher enzyme content. The enzyme was purified from the leaves by ammonium sulphate fractionation and most of the enzyme was precipitated at 30% ammonium sulphate concentration. From this investigation it becomes clear that optimum pH and temperature of the enzyme is 5 and 450C respectively. protein content of the sample was guantified according to the method of Lowry et al using bovine serum albumin as standard. Histochemical localization studies showed that acid phosphatase are mainly located in the bundle sheath regions.

    2) Project name : Screening of Region of Differences in Clinical isolates of Mycobacterium tuberculosis.
I have done this project at Mycobacterium research group, Rajiv Gandhi centre for biotechnology, Trivandrum under the guidance of Mrs. G. K Madhavilatha.
The annealing temperature for the primer pair of different RDs was
found by keeping a temperature gradient in the PCR from 60ºC to 72ºC. A 20µl PCR reaction was kept. After the standardization of annealing temperatures, DNA of clinical isolates was screened for the presence or absence of these regions i.e., to find out the presence of any polymorphism in these regions.
A total of 96 clinical isolates were screened to check for the presence of polymorphism in the RD regions 1, 6, 7, 8 and 10. Polymorphism was seen RD6, RD7, RD8 and RD10 but not in RD1.
