Description My project involved the isolation of Taq DNA Polymerase gene from a phage vector and its cloning in to Bacterial expression vector. Taq was isolated by PCR amplification. The amplified product was directly ligated into pUB-S-X plasmid. PUB-S-X is a bacterial expression vector which has ProU osmotic regulated promoter and sequences that are required to generate -T ends. PCR amplified Taq was directly ligated with linearized
pUB-S-X with –T overhangs. The recombinant clones generated were analyzed for the orientation of gene by colony PCR.
