Title of Master research thesis :
Water quality study of Buzzard’s Mouth creek and to evaluate the Cytotoxicity of organics and heavy metals extracted from creek sediment by using the MTT bioassay on BRL-3A cell line.
Abstract :
This study was aimed to assess the quality of water within the creek and identify the levels of heavy metals and nutrients in the creek water and sediments. A field study was conducted (13th of July 2010) to develop a data-base on physico-chemical parameters, heavy metals, and nutrient load in the water and sediments of the Buzzard’s Mouth Creek which passes through the barking riverside development site, East London. Eleven samples for water analysis and 8 for sediments analysis were collected from the Creek. Physico-chemical parameters were determined in situ by using YSI multiprobe whereas the concentration of nutrients were determined by using flow injection analyser (Lachat Quickchem 8500) and heavy metals were determined by using X ray fluorescence analyser (Twin-X, oxford instrument) and by Inductive couple plasma atomic emission spectrometry (ICP-AES). Cytotoxicity of organics and heavy metals extracted from creek sediment by using the MTT bioassay on BRL-3A cell line. All (except dissolved oxygen) physico-chemical parameters assessed in this study were within the recommended standard (EPA, 2010). The nutrient load of creek water was found in the range of phosphate (0.02 – 3.26 ppm), Nitrate/Nitrite (0.0009 – 0.1260 ppm), and Ammonia (0.0 – 3.74 ppm). Levels of heavy metals (Al, Fe, Ba, and Mn) were extremely higher in the Creek sediments as compare to water. This result suggests that even though the concentrations of phosphate and some of the heavy metals were higher, the water quality of the Buzzard’s Mouth Creek is suitable to maintain biodiversity and aquatic life which ultimately support to preserve the population of water vole.
