    Project name : Structure based drug design for hepatitis B.

    Description :
    The study of drug design is the approach of finding drugs based on their biological targets and inhibits the behaviour of disease related proteins. In hepatitis B virus the envelope protein are involved in viral binding and release into susceptible cells. It consists of core of double stranded DNA, a specific DNA polymerase and structural protein. Core antigen is localized predominantly with in the nucleus of infected hepatocytes.

    Accomplished
    :
    The main objective of the project was to design a drug compound to inhibit the core protein. The nucleotide composition, open reading frame, predicted peptide regions and primers were known through genomic approach. The stability of the protein sequence and physio-chemical nature of protein were studied. The protein was modeled using SPDBV with 80% accuracy, the protein was checked using procheck the potential active site was also been predicted using molecular cavities based on energy and surface area. Eight potential active amino acids were considered to be the best active site for targeting the protein and 14 similar drug molecules were taken for screening. The Tacrin molecule was found to be the best drug to inhibit the action of the protein. The protein ligand interaction was performed using Argus lab engine with flexible docking .finally Tacrin molecule with lowest free energy and high affinity was found to be best inhibitor for core protein.
