Title : Role of nitric oxide system in the pathophysiology of Sickle Cell Disease
- Considering the importance of nitric oxide (NO) in vasculopathy, my postdoctoral research involves nitric oxide synthase (NOS) expression, NO bioavailability and reactive oxygen species (ROS) generation from blood of steady state Sickle Cell Anemia (SCA) patients and SCA patients on hydroyurea (HU) therapy. Moreover, alteration in substrate (arginine) and cofactor (BH4) availability for NOS enzyme and underlying reactive oxygen- and nitrogen-induced events that contribute to SCA-induced vasculopathy was explored.
- In addition, supplementation of BH4, arginine, and drugs that increase NO bioavailability will be studied for the treatment of the disease in sickle cell disease transgenic mice.
