Process Engineer - Research & Development and Project Division
    Chief Role :
Handled development of products like Prebiotic : Fructo-oligosaccharides (FOS), Extraction of steviosides, a Sugar free from Stevia rebudiana plants in technical collaboration with CFTRI - Mysore.
Overseen the flask scale production of Fructo- oligosaccharides (FOS) syrup (upto 55% FOS content), scale-up studies at pilot scale production, Partial purification of enzyme catalyzing FOS production, Enzyme assay and analysis over HPLC, Immobilization of enzyme catalyzing FOS production and study of enzyme kinetics.
Development of high content Fructo-oligosaccharides (HCFOS, upto 95% FOS content).
Executed scale-up studies, conducting experimental trials as per guidelines, estimating process variables and data management.
Worked on equipment like Atomic Absorption Spectrometer, Flame Photometer, HPLC, Ion Exchange Chromatography, Fermenter, Refractometer, UV-Spectrophotometer and Gas Chromatography, Single effect Evaporator, Flash Evaporator, etc.
Accountable for preparing experimental protocols and reporting in response to development needs.
Testing and investigating new technologies and implementing on existing processes.
Communicating with Scientists, Technical Consultants and Dealers of various institutes, design firms and local equipment suppliers.
Troubleshooting various in-house production activities of ethanol production.
    Achievements :
Successfully optimised the :Steviosies extraction at lab scale.
High content FOS production at lab scale.
Improved the quality of FOS by reducing the microbial count of FOS by :
sterilisation of sugars before going for enzymatic reactions and
Membrane filtration (nominal purification) of enzyme; thus reducing the microbial count as per recommended and this suggestion has been adopted.
Analysed and found the one of possible cause may be clumping of yeasts as in ethanol fermentation unit there was reduction in productivity.
As mechanical alteration for fermenters to get proper mixing was not economic and possible for existing lakhs of capacity fermenters so thus suggested to use the dilute sulphuric acid that avoided the clumping and productivity has been increased upto some extent more than previous one. This suggestion been adopted.
Suggested going for continuous process rather than batch by immobilising the enzyme for FOS production. Thus by this way we will be able to use same enzyme for more than one batch and it would not alter the taste of our final product also.
In case of steviosides extraction from Stevia, Suggested the use of reverse osmosis process in between the solvent extraction and column chromatography will remove the unneeded amount of water and can solve the current existing problem.

    PROJECTS HANDLED :
    M.Tech. Curriculum
Title : Production of Substrate Specific Aminopeptidases from Lactic Acid Bacteria
    Period : Jul’08-Apr’09
Guide : Dr. K. Madhavan Nampoothiri, Scientist E1, Department of Biotechnology, National Institute for Interdisciplinary Science and Technology, (CSIR), Trivandrum
Synopsis : Newly isolated Lactic acid bacteria (LAB) as well as those procured from culture collection centres were screened for the production of various substrate specific aminopeptidases. Among all the strains screened, L. brevis (NRRL B-1836) was found to produce quantifiable amount of intracellular arginine specific aminopeptidase (EC 3.4.11.6). Attempts were made to formulate the production medium (supplementation of carbon and nitrogen sources) and the inoculum size to enhance the production yield. The productivity of arginine-aminopeptidase in 5L fermentor scale up was calculated to be 36 IU/L/h. The Luedeking and Piret model was tested for intracellular production of aminopeptidase and the data seemed to fit well, as the correlation coefficient was 0.9964 for MRS. The aAP and ßAP was 0.4865 and 0.0046 respectively in MRS medium indicating that the yield was predominantly depended on growth. The organism produces lactic acid and was also found to tolerate pH 2.0 -3.0 and 0.3- 0.5% bile salts, the most important probiotic features. The partial purification of enzyme, arginine specific aminopeptidase has been done by ultrafiltration and gel filtration chromatography by which the enzyme activity was found to increase by 3 folds. The molecular weight of the enzyme, arginine specific aminopeptidase has been determined by Gel Electrophoresis: SDS-PAGE and it was found to be in between 40 to 50 kDa.
Title : Trace and Bulk Salt Analysis of Mammalian Cell Culture Media
    Type : Industrial Training Programme
Institute : Biocon Pvt. Ltd., Bangalore, Mammalian Cell Culture Lab, Department of Research and Development at Period : May’08-Jul’08
Synopsis : Mammalian cell culture is technically more complex and more expensive than microbial cell fermentation. Mammalian cells are only used in the manufacture of therapeutic proteins that show extensive and essential post-translational modifications. Animal cells are often more advantageous than yeast or bacterial cells for the production of recombinant proteins on a large scale. The media used for the fermentation process contained a carbon source, nitrogen source, basal salts and trace elements. The amount of therapeutic protein produced during the fermentation process was analysed by High Pressure Liquid Chromatography (HPLC) by using Agilent 1200 & 1100 series. The media was also analysed at regular time intervals for the amount of anions and cations present by Ion exchange chromatography using Dionex ICS-3000.

Title : Bioremediation: Microbial Degradation of Plasticizers
Guide : Dr. B.G. Pujar, Head of Department, Department of Biotechnology, Bapuji Institute of Engineering & Technology, Davangere.
Synopsis : Phthalate and phthalate esters (plasticizers) are most frequently encountered as environmental pollutants due there large scale use in industries and agriculture for various purposes. Toxicity of these chemicals to human and animal system has been reported. Chemical degradation of these compounds can take six to hundreds of years. My investigation was undertaken to know the biodegradative pathway of dimethyl and diethyl phthalate in comparatively less time and isolated Pseudomonas strains of bacteria from soil and studied their biodegradative pathway. The morphological, biochemical and physiological characterisation of organism has been done for tentative identification and also the isolation and characterisation of metabolites has been carried out.
