- A project on “Effect of Tumour Microenvironment on Bone Marrow derived Dendritic cells” under the guidance of Dr. Bhavani Shankar, S.O-F in Bhabha Atomic Research Centre (BARC) (3 months).
Abstract : Dendritic cells are professional antigen presenting cells in immune system. Dendritic cells are capable of capturing antigens, processing them and presenting them on the cell surface along with appropriate co-stimulatory molecules. In case of cancer, tumor derived factors induce dendritic cell dysfunction and alter the differentiation, maturation, and functional activation of dendritic cells as a mechanism for immune suppression. In this study we have observed that when bone marrow derived dendritic cells (BMDC) were cultured in presence of Tumor conditioned medium (TCM) obtained from EL4 T cell lymphoma and B16F10 cancer cell lines, the expression of the co-stimulatory molecules CD 40, CD80, CD86, MHC II and CD11c is suppressed drastically. Secretion of IL12 and TNFa cytokines by BMDC were also analyzed in the presence of TCM during different periods of BMDC development. These cytokines secretion were also inhibited by TCM. However, when TCM was present only during maturation, the suppressive effect was not seen. Agents like LPS induce maturation of dendritic cells. We observed that LPS stimulation is able to over-ride the suppressive effect of TCM and restore the expression of co-stimulatory molecules and MHC.
