Topic : Cloning & Expression of Brugia malayi Hexokinase
Techniques involved : PCR, Plasmid and Genomic DNA isolation, Preparation of
competence cell and transformation techniques, Restriction
enzyme analysis, SDS-PAGE, Native-PAGE, Affinity
chromarography.
Hexokinase being the regulatory enzyme of glycolytic pathway therefore may be a potential chemotherapeutic target. The complete ORF of the hexokinase of Brugia malayi was amplified by PCR. The PCR amplified product was cloned in cloning vector TOPO-TA and ligated mixture was transformed in competent DH5a. Expression was checked in pTriEx-4 and the gene was successfully expressed in this vector.
