    # Immunological Experiments from Edu biotech, Bangalore.
    20 Days
    This project was for a duration of twenty days
    The project is all about the different types of immunological experiments like Enzyme Linked Immuno Sorbent Assay (ELISA) and the different types of Antigen-Antibody reactions.
    # Sub-cloning of CP gene & Protein related experiment from Indo-American Hybrid Seeds (IAHS), Bangalore.
    2 Months
    The project is on the sub-cloning of coat protein gene.
    Here the coat protein gene present in a virus was extracted using different RDT methods like plasmid isolation, agarose gel electrophoresis restriction digestion and gel elucidation and then ligated with a vector (pGPTV,a binary vector) the next step was competent cell preparation and transformation to transform the plasmid containing CP gene after this comes the synthesis of transgenic plant containing the coat protein gene using the plant tissue culture technique which resulted in a virus resistant plant. The plant chosen was tomato.
    In the first phase, the selected clones were taken to carryout sub cloning and Agrobacterium mediated transformation of CP gene in tomato plants. The developed transgenic plants were evaluated through PCR.
    The second phase included detection of ToMV infection in different plants by molecular techniques such as SDS-PAGE Immunodetection and ELISA.
    # Immunosenescence (internal project)
    1 Month
    This is an internal project conducted for a month. It deals with the senescence of the immune system. Immunosenescence refers to the gradual deterioration of the immune system brought on by natural age advancement. It also explains the causes of immunosenescence, the cure isn’t clear because it’s a topic under study.
    # Novel Therapeutic Approaches To Cancer Treatment By Inhibition Of DNA Damage Signaling Pathways (Biomed Informatics, Medwin Hospital, Hyderabad)
    1 ½ months
    Cell signaling pathways are fundamental to the understanding of cancer. A complicated series of signals tell the cells when to divide and when to stop growing. When the signaling pathways are working correctly, the spoiled cell recognizes that it is defective and either repairs the damaged DNA or triggers apoptosis. This project addresses a side of clinical research starting from raw genome data till drug design and by making use of different computer programs we availed on the wealth of information available in public databases and biological literature. The goal of this project work is to identify the implication of DNA signaling pathway in cancer cells. We held a particular focus on erbB2 protein, its way of relaying proliferation signals to other TKs and imparting resistance to apoptosis in tumors. Homology modeling is a major part of this project work, we modeled the protein erbB2 and identified its active sites and addressed some of the available anti-cancer drugs.
    The project consists of three different parts :
    Genomics
    Proteomics
    Drug Designing
    In the genomics part of the project the sequence of the ERBB2 gene and the Open reading frames where found using the different NCBI tools like ORF finder, GENSCAN etc.The BioEdit software which provides the basic functions for proteins and nucleic sequence editing, alignment manipulation and analysis was used. Sequence Alignment was done using BLAST for similarity search, pair wise and multiple sequence alignment was also done. Genomics part of the project explained a vast amount of information which links gene activity with diseases.
    The proteomics part covers many areas like :
    Structure prediction and protein modeling(Primary, Secondary and Teritary)
    Function prediction
    Protein folding and topology
    Active site analysis
    Target identification and optimization
    Protein targeting
    The different computational tools and software used in this part of the project was :
    Swiss PDB Viewer, BioEdit, PovRay etc.
    Based on the active sites present drug was designed in the drug designing part of the project and the different programs used here is :
    Argus lab, Vegazz, ChemDraw Ultra, Cache, HyperChem etc. The drugs which binds with the active site more accurately are :Lapatinib, Femara, PKI-166, HKI-272
