    (1) General Position Objective

    Responsible for
    (1) Contributing in Developing and launching new Recombinant Antibody Products.
    (2) Set up and run high throughput assays on antibody screening.

    RESEARCH EXPERIENCES

    MSc Project: “Analysis of the Interaction Between hRev7 and hRev3 in Mammalian Cell Cycle”
    Subunits of DNA polymerase - were cloned in bacterial strains and transfected into HeLa cells and their interactional function was studied up on DNA damage using immune fluorescence microscopy. Truncated mutants of these genes were synthesised for identifying the binding loci. His tag Mad2B protein was synthesised and purified for NMR studies.

    B. Tech Project: “Algal Fuel Cells and Electricity Generation”
    Nostoc, Westiellopsis and Anabaena were grown in specially designed unit and the electron flow was estimated by multimeter, the current generation persisted for 4 days. Cell membrane Cytochrome C of these algal species was purified by affinity chromatography and the concentration was estimated by HPLC. Nostoc with more Cytochrome C in its cell membrane produced more electricity.

    Research Experiences


    1. Associate Research Fellow in the Research Centre of LifeCell International Private Limited, Chennai. I have experience in the area of Stem cell and Regenerative Medicine for more than four years (since January 2006).

    Job Responsibilities:

    a. Isolation, Characterization, Maintenance and Cryopreservation of Mesenchymal stem cells from human adipose tissue, cord blood, cord lining and placenta. And carried out isolation, expansion and colony forming unit assay of human haematopoietic stem cells from Umbilical cord blood. In addition, I handled Magnetic Associated Cell Sorter for haematopoietic stem cells.

    b. Involved in validation of protocol for Isolation, Culturing, large scale expansion and Cryopreservation of Mesenchymal stem cells from human cord lining and menstrual blood for commercial purpose and the expansion technology has been launched currently. In addition, have experience in differentiation study on cord lining mesenchymal stem cells into Osteocytes, chondrocytes, Adipocytes, human lung airway epithelial cells and bronchial epithelial cells.

    c. Validation and Implementation of Mycoplasma and Endotoxin testing (both Gel clot and Kinetic method) for routine Quality control of chemicals and reagents.

    d. Also involved in development of infrastructure for the Research Unit and processing units expansion and technical support to the marketing department .

    e. Preparation of the annual budget for R&D projects and its cost breakups. Projecting the cost analysis of the technical protocols and procedures which is validated for commercial launch and quality control operations.

    ABSTRACT: Standardization of the stem cell isolation and characterization procedure from Amniotic sac and Chorionic villi and compared in terms of cell morphology, population doubling time and cell surface markers and the same was done in three different cell culture media for comparison.


    3. M.Sc. APPLIED MICROBIOLOGY PROJECT - entitled “Pleurotus sajor-caju, the oyster mushroom as the source of Chitin” at the PSG College of Arts and Sciences, Bharathiar University, Coimbatore, India.

    ABSTRACT: Polymeric chitin was isolated, purified and quantitated. In addition, comparitive analysis of fungal mass generation in different concentrations of nitrogen and carbon growth media.

    4. B.Sc. PROJECT entitled as “Development of polyclonal antibody based latex agglutination test against the fish pathogen Aeromonas hydrophila using thermo stable somatic “O” Antigen” at The American College, Madurai Kamaraj University, Madurai, India.

    ABSTRACT: Polyclonal antibodies raised in Sheep against heat inactivated A. hydrophila antigen are isolated from its sensitized serum by salt fractionation method and quantitated with spectral method. Furthur the quantitated antibodies are coated on the latex beads which can give visible agglutination when live A. hydrophila bacterial cells are added in a slide. The pivotal logic in this project is the heat inactivation of the bacteria leads to loss of thermolabile antigens which is common to all species and only thermo stable antigens will remain which is not common for all other species (i.e, species specific) that ensued in avoiding the cross reaction with other Aeromonas species.

Immunology and Microbiology: Basic Microbiological techniques and antimicrobial activities, polyclonal antibody generation in laboratory animals, handling laboratory animal, immunodiffusion test and H- serotyping. The present study was aimed to investigate the effect of Nigella sativa oil (1ml/kg b.w.) in vivo antioxidant studies and biochemical parameters of kidney in presence and absence of cisplatin toxicity in rat and to study the oral administration of Nigella sativa (L) (500mg/kg b.w.) for a week in rats.

    Thesis Title : Evaluation of extracts of some marine molluscs on bone metabolism: An Experimental Study.
    Bone loss occurs when there is an imbalance between bone resorption and bone formation, resulting in increased risk of fracture leading to a skeletal disorder termed as OSTEOPOROSIS. OSTEOIMMUNOLOGY is an interdisciplinary research field combining the exciting fields of osteology and immunology. In this study, marine molluscs Turbo brunneus and Euchelus asper were found to be effective in inhibiting bone resorption produced experimentally in mice by bilateral ovariectomy. Turbo brunneus was able to inhibit the bone resorption by the regulating the T cell function. Turbo brunneus also proved to be more effective in inhibiting bone resorption by suppressing osteoclasts and also the cytokines involved in the osteoclastogenesis. Hence, it was confirmed that Turbo brunneus can prove to be good candidate for the development of an antiosteoporotic drug.

    Title of the project : Characterisation of Bioactive Compounds from Marine Mollusc and Porifera that regulate Osteoclastogenesis.
    Post menopausal osteoporosis induced by estrogen deficiency by far is the main cause of bone resorption in elderly women. Using an ovariectomised mouse model of estrogen deficiency we observed that the oral administration of some marine extracts to ovariectomised mice could reduce the increased bone resorption. This reversal of bone resorption was also confirmed by Raman spectroscopy studies which demonstrated an increase in the mineralisation of the bones of mice treated with these marine extracts. Immunophenotyping studies revealed the oral administration of the marine extracts reduced the elevation of various T cell markers. Our data revealed that the marine extracts could drastically reduce the TNF, IL-6 secretion which showed that the extract is capable in actively regulating osteoclastogenesis. These extracts were also able to reduce the RANKL induced resorption pits formation by the osteoclast on artificial calcium coated slides.
